摘要
目的探讨转录因子(calcium-responsivefactor,CaRF)在大鼠皮肤创面愈合中的作用。方法建立大鼠背部皮肤全层切割伤模型,随机将大鼠分为3组盐水对照组,CaRF过表达处理组,CaRFRNAi处理组,以脂质体转染的方式将CaRF和CaRFRNAi导入创面组织,观察3组大鼠背部皮肤伤口愈合状况。结果CaRF过表达组大鼠皮肤愈合快,愈合质量好,CaRFRNAi组大鼠皮肤愈合最慢,愈合质量差。结论CaRF促进大鼠皮肤创面愈合,为临床创面愈合提供了新的思路。
Objective To investigate the roles of calcium-responsive factor (CaRF), a transcription fac- tor, in wound healing. Methods An excision of 1cm×1cm full thickness wound were inflicted on the backs of SD rats, and then the rats were randomly divided into 3 groups, saline control, CaRF RNAi treatment, CaRF over-expression treatment, with the animals of later 2 groups receiving CaRF or CaRF RNAi by lipofectin trans-fection. The progress of wound healing was observed by wound healing index, morphological changes, contents of hydroxyamino acid and total protein in granulation tissues, and the expression of CaRF at the 3rd, 7th, 14th day post-injury respectively. Results CaRF RNAi treated group showed slower wound healing while the healing in the CaRF over-expressed group was faster and better with an extra reduction of 28.6 % in wound as compared to control groups. Oonclusion The transcription factor CaRF may be involved in wound healing, which may provide a foundation for the development of new treatment strategy for wound healing after skin injury.
