摘要
目的研究能量可控陡脉冲治疗兔移植性肿瘤的最佳剂量及安全性。方法建立10只新西兰大白兔VX2移植性乳腺癌模型,通过正交设计给予ECSP治疗,TTC染色观察其治疗体积情况,通过直观分析寻找最佳剂量。建立20只新西兰大白兔VX2移植性肝癌模型,给予ECSP治疗后取其周边正常组织,行DNA片断化分析及彗星实验。结果在ECSP的4个参数中,电压对治疗效果的影响最大。在不考虑各因素间相互影响的条件下,电压为150V,频率为100Hz,脉宽为0.01μm,作用时间为15min时的治疗效果最理想。ECSP治疗后对周边正常组织的DNA电泳为一完整条带,无明显彗星扫尾现象发生,表明无DNA损伤。结论ECSP治疗兔移植性肿瘤的最佳剂量为电压150V,频率100Hz,脉宽0.01μm,作用时间15min,对周围组织安全。
Objectives To explore the optimal dosage and evaluate pulse (ECSP) in the established in 10 New treatment of implanted rabbit tumor. Methods The the safety of energy controllable steep models of VX2 breast carcinoma were England rabbits, and then the animals were given ECSP treatment under orthogonality designed dosages of ECSP. The volumes of necrosis tumors were measured by TTC staining. The best dosage was analyzed by intuitionistic analysis. The models of VX2 liver carcinoma were established in 20 New England rab- bits. The normal tissues surrounding the tumor were extracted, the DNA fragmentation analysis and comet assay were carried out. Results Voltage took the priority among the 4 parameters, and for best dosage they were 150 V for voltage, 100 Hz for frequency, 0.01μm for pulse width, and 15 min for time if the interaction of these 4 factors was not considered. The electrophoresis of DNA in the peripheral normal tissue was an integrity straining after ECSP treatment. No comet tailing was observed, indicating no DNA lesions after ECSP treat- ment. Conclusion The best dosage is 150 V for voltage, 100 Hz for frequency, 0.01μm for pulse width and treating for 15 min. The ECSP is verified to be safe.
