腰神经根受压模型大鼠神经根局部炎症因子对温和灸的反应 被引量:6

摘要 目的:观察温和灸对腰神经根受压模型大鼠受压神经根局部炎症因子白细胞介素1和白细胞介素6的调节作用,并与灌胃给药莫比可相比较。方法:实验于2005-09/2006-04在上海第六人民医院实验中心完成。①分组:将40只SD大鼠随机分为4组,正常对照组,模型对照组,温和灸组及莫比可组,每组10只。②方法:温和灸组、莫比可组、模型对照组通过手术建立大鼠腰神经根压迫模型,正常对照组做与以上各组相同的捆绑固定,不予手术。温和灸组穴位选取受压神经根相应节段右侧的夹脊穴,采用精制清艾条以穴位为中心、半径为5mm的区域进行温和灸,20min/次,1次/d,7次为1个疗程,共治疗14次。莫比可组按3.75mg/kg灌胃莫比可(10mL生理盐水含药物5mg),每天灌胃1次。正常对照组及模型对照组:生理盐水灌胃10mL/kg,每日灌胃1次。于治疗14d后麻醉下处死取受压神经根组织。③评估:于光镜下观察受压神经根组织形态学变化;用免疫组织化学技术检测神经根局部炎症因子白细胞介素1、白细胞介素6水平。结果:40只大鼠均进入结果分析。光镜下观察温和灸组神经节细胞、神经纤维结构基本正常,见极少量炎细胞。与模型对照组相比,温和灸组与莫比可组神经根局部炎症因子白细胞介素1、白细胞介素6的含量有不同程度的减少(P<0.05);温和灸治疗组白细胞介素1、白细胞介素6的水平低于莫比可组(P<0.01)。结论:温和灸及西药莫比可均能有效抑制神经根局部炎症因子分泌,其中温和灸作用迅速,其效果明显优于莫比可。 AIM: To study the effect of warm moxibustion on nerve root local intedeukin-6 (IL-6) in rat model with lumbar nerve root compression perfusion. inflammatory factors: interleukin-1 (IL-1) and and compare with Meloxicam tablet by gastric. METHODS: The experiment was completed in the Ex.periment Center of Shanghai Sixth People's Hospital from September 2005 to April 2006.①Grouping: 40 SD rats were stochastically divided into 4 groups: normal control group, model control group, warm moxibustion group and Meloxicam tablet group, there were 10 rats in each group.②Methods: Model control group, warm moxibustion group and Meloxicam tablet group adopted rat lumbar nerve root compression by surgery for constructing model. The rats of normal control group received the same colligation and fixation as above groups, and did not give the surgery. The acupuncture point of warm moxibustion group was selected as Jiaji (Extra) points in the right side of compressed nerve root. Warm moxibustion was carried on a region taking acupuncture point as the center and the radius was 5 mm by purified moxa roll, 20 minutes once, one time per day, 7 times as 1 treatment course, and altogether for 14 times. Meloxicam tablet group was treated by gastric perfusion at the dose of 3.75 mg/kg (10 mL physiological saline included 5 mg medicine), once every day. The normal control group and model control group were treated by gastric perfusion of 10 mL/kg physiological saline, once every day. After 14 days of treatment, the compressed nerve root was taken when rats were anaesthetized to death.③Evaluation: Under the light microscope, the change of tectology of the compressed nerve root was observed, IL-1 and IL-6 were tested by immunohistochemistry technique. RESULTS: All the 40 rats entered the result analysis. The structure of ganglion cell and the nerve fiber was normal, and the extremely few inflammation cells were observed under the light microscope in warm moxibustion group. Compared with the model control group, the content of IL-1 and IL-6 in the warm moxibustion group and the Meloxicam tablet group were reduced at the varying degrees (P 〈 0.05). The content of IL-1 and IL-6 in the warm moxibustion group was lower than that of Meloxicam tablet group (P 〈 0.01). CONCLUSION: Both the warm moxibustion and the Meloxicam tablet can effectively inhibit the exudation of the local inflammation factors from the compressed nerve root. Warm moxibustion, presenting fast action, is superior Meloxicam tablet.
