摘要
目的:探讨HOXa-10基因在小鼠胚胎着床过程中的作用。方法:分别应用FQ-PCR和免疫组化方法检测未孕及早孕第1、2、3、4、5、7天小鼠子宫内膜HOXa-10基因及蛋白的表达。结果:FQ-PCR测得随着小鼠妊娠天数的增加,子宫内膜HOXa-10基因表达量也逐渐增高,在孕4天达到峰值免疫组化分析显示HOXa-10蛋白在子宫内膜的表达规律与FQ-PCR结果一致。结论:在小鼠胚胎着床过程中,HOXa-10在子宫内膜中高表达,可能对子宫内膜容受性变化有重要的调节作用。
Objective:To investigate the effect of HOXa-10 gene in the mouse uterus during embryo implantation. Method: real-time fluorescence quantitative PCR (FQ -PCR) and immunohistochemistry were used to detect the HOXa-10 gene and protein of the mouse of un-pregnant pregnant d1,d2,d3,d4,d5 and d7. Result:The HOXa-10 mRNA/β-actin mRNA expression in pregnant group is higher than that of non-pregnant group, and the results showed gradual hoist trend as days past, the HOXa-10 mRNA expression reached the maximum at pregnant d4. Immunohistochemical analysis showed the same results as FQ-PCR. Conclusion:HOXa-10 is persistently expressed in mouse endometrium during the early pregnancy and maybe participates in the regulation process of mouse blastodyst implantation.
