Asymmetric cell division is a fundamental pattern regulating cell differentiation during animal development and is also conserved in evolution. A polarized progenitor gives rise to two daughter cells with different fates through the asymmetric division which involves the asymmetric distribution of cell fate determinants and the orientation of the mitotic spindle. Both processes need the presence of certain cell polarity. In this paper, how PAR/aPKC complex, a highly conserved protein complex, functions in the establishment and maintenance of cell polarities as well as in the regulation of deterninants distribution and mitotic spindle orientation were reviewed, and some recent findings and progresses in this field were also discussed.
