Application of traditional Chinese medicine in stem cell transplantation for cardiovascular diseases

学术背景:在骨髓干细胞移植治疗心脏疾病的研究中,常用的移植方法是将干细胞直接注射到受损的心肌,可促进心肌组织修复的外周血干细胞数量的增加可通过中药骨髓干细胞动员剂实现。目的:总结中医药在干细胞移植治疗心血管疾病中作用的研究进展。检索策略:由该论文的研究人员应用计算机检索Pubmed数据库1995-01/2003-12的相关文献,检索词"stemcells,transplantation,traditionalChinesemedicine,cardiovasculardisease",并限定文章语言种类为English。同时检索万方数据库2003-01/2007-12的相关文章,检索词为"干细胞移植,中医药,心血管疾病"。共检索到62篇文献,对资料进行初审,纳入标准:①与中医药在干细胞移植治疗心血管疾病中作用密切相关。②同一领域选择近期发表或在权威杂志上发表的文章。排除标准:重复性研究。文献评价:文献的来源主要是中医药对干细胞移植治疗心血管疾病干预作用的文章。所选用的26篇文献中,5篇为综述,其余均为基础实验研究。资料综合:中药在干细胞增殖及向心肌细胞分化的研究中有了突破性的进展,特别是中药的单味成分及复方制剂的提取物在动物实验中都有理想的结果及显著疗效。进行中药诱导胚胎干细胞分化为心肌细胞的探索,将为应用中医药治疗心肌梗死提供新的理论依据。中药具有临床应用安全和长期有效的特点,能扩张冠状动脉,抗心肌缺血;能调节机体免疫,抗排斥反应;能促进细胞增殖,抗细胞凋亡;能增强基因调控,促进蛋白表达等。结论:中药干预的研究虽然起步较晚,目前也仅是体外实验研究阶段,不管是复方制剂,还是单味药提取的有效的药物成分,在体外培养都脱离了人体环境,实验结果有其局限性。骨髓干细胞与中药诱导分化将可能成为中医药走向现代化的一个标志,具有重大的实际意义。

BACKGROUND： In bone marrow stem cell transplantation for heart diseases, stem cells are directly infused into injured myocardium. Increased amount of peripheral blood stem cells, which can accelerate the repair of myocardium, can be achieved by traditional Chinese medicine bone marrow stem cell mobilization agent. OBJECTIVE： To summarize the research progress of traditional Chinese medicine in stem cell transplantation for cardiovascular disease. RETRIEVE STRATEGY： The Pubmed Database was undertaken to identify relevant literatures published from January 1995 to December 2003 with the key words of ＂stem cells,transplantation, traditional Chinese medicine, cardiovascular disease＂ in English. Simultaneously, the Wanfang Database was undertaken to identify relevant articles published from January 2003 to December 2007 with the same key words in Chinese. Totally 62 articles were retrieved. After primarily searching, inclusive cdtena included ①articles on close correlation with traditional Chinese medicine in stem cell transplantation for cardiovascular disease. ②Of the articles on the same circle, articles published on authoritative joumals or recently published. Exclusive critena included articles with repetitive research. LITERATURE EVALUATION. The articles were mainly on traditional Chinese medicine in stem cell transplantation for cardiovascular disease. Of the 26 articles, 5 were review, and others were basic experimental research. DATA SYNTHESIS： The application of traditional Chinese medicine in stem cell proliferation and differentiation into myocardial cells has outstandingly developed. In particularly, single component and extractive of compound Chinese herb can have significantly curative effects in animal tests. Researches on embryonic stem cells differentiating into cardiomyocytes induced by traditional Chinese medicine will offer new evidences for the application of traditional Chinese medicine in the treatment of myocardial infarction. Traditional Chinese medicine is characterized by safety in clinic and long-term effect. It can prevent myocardial ischemia by expanding coronary artery, prevent rejection by adjusting body immunization, prevent apoptosis by accelerating cell proliferation and enhance protein expression by enhancing gene regulation. CONCLUSION. The study on traditional Chinese medicine is late, and it is only in vitro expenment at present. Studies on either compound preparation or effective drugs of single drug extractive are away from human environment, and the results of the experiment are not full. Bone marrow stem cell differentiation with traditional Chinese medicine can be a mark of the application of traditional Chinese medicine in modem studies.
