肝细胞生长因子抑制AngⅡ诱导心肌成纤维细胞增殖和胶原合成

摘要 目的检测肝细胞生长因子(HGF)对血管紧张素Ⅱ(AngⅡ)诱导的SD大鼠心肌成纤维细胞(CFbs)增殖和胶原合成的影响,探讨其在高血压心室重构中的作用。方法用消化法培养CFbs,以HGF和(或)AngⅡ刺激CFbs,以蛋白免疫印迹方法分析CFbsⅠ型胶原蛋白合成,MTT法检测CFbs的增殖。结果AngⅡ促进CFbs增殖和Ⅰ型胶原蛋白的合成,HGF可抑制这些作用。结论HGF能抑制AngⅡ诱导的SD大鼠CFbs增殖和心肌胶原沉积,从而减少AngⅡ引起的高血压心室重构,实现保护心脏的作用。 Objective To investigate the effects of hepatocyte growth factor (HGF) on proliferation and collagen synthesis induced by angiotensin Ⅱ ( Ang Ⅱ) in SD rat cardiac fibroblasts (CFbs) , and determine the role it plays in hypertensive ventricular remodeling. Methods CFbs in SD rat were cultured in vitro and treated with different concentrations of AngⅡ and/or HGF. Collagen type I synthesis of CFbs was measured by Western blotting, and proliferation of CFbs was detected by MTT assay. Results Ang Ⅱ promoted collagen synthesis and proliferation in CFbs in a certain concentration range, which can be significantly inhibited by HGF. Conclusion HGF can inhibit the proliferation and collagen synthesis induced by Ang Ⅱ in SD rat CFbs, and may protect against hypertensive ventricular remodeling.
