摘要
目的建立稳定的胆道外引流和胆汁收集的大鼠原位肝移植模型。方法在"双袖套法"技术基础上,引入大鼠躯干"马甲"式外固定器保护胆道引流管和胆汁收集装置。结果大鼠胆道外引流原位肝移植手术60次,手术成功率93.3%(56/60)。术后1d、1周和4周存活率分别为86.7%(52/60)、76.7%(46/60)和68.3%(41/60)。因胆瘘和胆道梗阻死亡各3例(10.0%)。结论本大鼠肝移植模型胆道外引流稳定,胆道并发症发生率低,可为肝移植后胆汁生物学分析研究提供技术支持。
Objective To develop a rat model of orthotopic liver transplantation with stable biliary extra-drainage and bile collection. Methods The model was established with "two cuff" technique and introduction of waistcoat-like external fixator for the trunk of the rat, which was designed to protect the biliary extra-drainage tube and bile collection apparatus from destruction by the rat. Results Sixty cases of rat orthotopic liver transplantation were performed, and the rate of successful operation was 93.3% (56/60). The survival rate on the post-transplant first day was 86.7% (52/60) , and the survival rates of post-transplant 1- and 4-week were 76.7% (46/60) and 68.3% (41/60) , respectively, Three cases died of biliary fistula and 3 died of biliary obstruction ( 10.0% ). Conclusion For stable biliary extra-drainage and low incidence rate of biliary complications, this rat model of orthotopic liver transplantation is reliable for those studies involving bile analysis after liver transplantation.
