摘要 目的观察蒿甲醚(ART)在体外对人胰腺癌SW-1990细胞的杀伤作用和对细胞增殖和凋亡的影响,以及在体内对荷瘤裸鼠移植瘤的抑制作用。方法采用MTT法检测ART对SW-1990细胞的杀伤作用;采用流式细胞仪检测ART对SW-1990细胞周期和凋亡的影响。建立人胰腺癌裸鼠移植瘤模型,通过测量裸鼠体质量、移植瘤瘤径和瘤质量,计算肿瘤体积和抑瘤率,观察ART在体内的抗肿瘤活性。结果MTT结果显示,ART对SW-1990细胞的杀伤作用与时间-剂量呈正相关(P<0.05);流式细胞仪检测显示,ART使SW-1990细胞阻滞于G0/G1期,并诱导细胞凋亡;荷瘤裸鼠体内实验显示,中、高剂量的ART(4、8 mg)对裸鼠移植瘤的生长抑制效果显著(P<0.05),抑瘤率分别为48.10%和53.51%。结论ART在体外及荷瘤裸鼠体内对SW-1990细胞有明显的抑制作用;其抑制作用与阻滞细胞周期和诱导细胞凋亡有关。 Objective To investigate the in vitro killing effect of Artemether (ART) on human pancreatic cancer cell lines SW-1990 and effect on cell proliferation and apoptosis, and explore the inhibitory effect on the growth of transplanted tumor in vivo. Methods The inhibitory effect of ART on SW-1990 cell lines was detected by MTT assay, and the effect on cell cycle and apoptosis was evaluated by flow cytometry. The pancreatic cancer model was established with tumor-bearing nude mice, and the in vivo anti-tumor effect of ART was observed by measuring body weight, tumor diameter and calculating tumor volume and tumor growth inhibition rate. Results It was indicated by MTT assay that the killing effect of ART on SW-1990 cell lines was positively correlated to time and dosage (P 〈 0.05). The result of flow cytometry demonstrated that ART treatment induced SW-1990 cell arrest in G0/G1 phase and cell apoptosis. In vivo experiment on tumor-bearing nude mice revealed significant anti-tumor effect in moderate and high-dosage ART groups(4 and 8 mg) (P 〈 0.05) with the inhibition rate of 48. 10% and 53.51% , respectively. Conclusion Artemether has significant inhibitory effect on SW-1990 cell lines in vitro and marked effect on tumor growth of tumor-bearing nude mice in vivo, which may be correlated to cell cycle arrest and cell apoptosis.
