摘要
以聚3-羟基丁酸酯(PHB)和聚乙二醇(PEG)共混物为原料,引入牛血清白蛋白(BSA)作为模型药物,采用复乳化溶剂蒸发法制备包埋BSA的PHB/PEG微球,研究了制备工艺对微球性能的影响.实验结果表明,较优化的微球制备工艺为:W1/O=1/5,O/W2=1/5,聚合物浓度为4%w/v,配比m(PI-IB):m(PEG)=4:1;该工艺所得微球的平均粒径在10μm左右,BSA突释率为17.8%,30d的累积溶出量达70%左右,释放特性比较理想.
Microspheres containing bovine serum albumin (BSA, a model drug) were prepared via double emulsion solvent evaporation method using compound of poly(3-hydroxybutyrate) (PHB) and polyethylene glycol (PEG) as the matrix. Effect of processing conditions on the properties of BSA-PHB/PEG microspheres were studied. The results show that improved processing conditions involve W1/O= 1/5 and O/WE=1/5. The concentration of polymer compound is 4% w/v with a 4 : 1 mass ratio of PHB and PEG. The mean particle size of the microspheres prepared under this conditions was about 10μm. It had a good release characteristic in vitro. Initial burst of BSA was 17.8% and accumulative BSA released after 30 d reached 70% around.
