摘要
目的探讨2-甲氧雌二醇对HL60白血病细胞增殖及凋亡的影响。方法运用四唑蓝(MTT)法和彗星电泳法检测2-甲氧雌二醇对HL60白血病细胞的增殖抑制及促凋亡作用;应用流式细胞技术检测Bcl-2蛋白的表达。结果(1)2-甲氧雌二醇在10~50μM浓度范围内作用72h以及30μM2-甲氧雌二醇在作用24、48、72h后对HL60白血病细胞有增殖抑制作用,这种作用呈时间-剂量依赖关系。(2)2-甲氧雌二醇在作用浓度为10、30、50μM时,HL60白血病细胞凋亡率分别为8.20%、28.20%、45.30%,与阴性对照组(1.00%)差异有统计学意义;30μM2-甲氧雌二醇在作用24、48、72h后,HL60白血病细胞凋亡率分别为3.71%、17.72%、43.58%,48、72h作用时间组与阴性对照组(1.00%)差异有统计学意义。(3)30μM2-甲氧雌二醇作用72h后的细胞,Bcl-2蛋白的表达较对照组显著减少。结论2-甲氧雌二醇具有对HL60白血病细胞增殖抑制及促凋亡作用,Bcl-2蛋白表达下调在此过程中可能发挥着重要的作用。
Objective To investigate the effect of 2-methoxyestradiol on the proliferation and apoptosis of HL60 cells in vitro. Methods After treated with 2-methoxyestradiol, the cell proliferation was detected with MTT, and apoptosis was measured with comet assay. Expression of Bcl-2 protein in HL60 cell was detected with flow cytometry. Results After treatment with various concentrations (10, 30, 50 μM) of 2-methoxyestradiol for 72h or 30 μM for different time-points (24, 48, 72 h ), the growth inhibition rate exhibited significant increase (P〈0. 05) in time-dose dependent manner; and the apoptotic rate accounted for 8.20%, 28.20%, 45.30%, 3.71%, 17.72%, and 45.38% respectively, Bcl-2 protein decreased significantly in 30 μM 2-methoxyestradiol 72 h group compared with that of control group. Conclusion 2-methoxyestradiol can inhibit proliferation and induce apoptosis of HL60 in vitro; down regulation of Bcl-2 protein may play an important role in the procedure.
