穹窿海马伞切割后海马内NSCs的增殖和向神经元的分化 被引量:6

Proliferation and Differentiation of the Endogenous Neural Stem Cells in Hippocampus into Neurons After Fimbria Fornix Transection

摘要 目的观察切割穹窿海马伞侧和非切割侧大鼠海马内自体神经干细胞的增殖和向神经元分化的情况。方法切割SD大鼠右侧穹窿海马伞,术后2 d腹腔注射BrdU,连续5d,于术后28 d取脑冰冻切片,进行BrdU/NF-200免疫荧光双标检测。结果切割穹窿海马伞侧海马内的BrdU免疫荧光阳性细胞明显多于非切割侧,且发现有BrdU/NF-200双标神经元,而非切割侧未见到BrdU/NF-200双标神经元。结论穹窿海马伞切割后,切割侧海马内自体神经干细胞增殖加快,并可向神经元分化。 Objective To investigate the proliferation and neuronal differentiation of the endogenous neural stem cells (NSCs) in the bilateral hippocampus of adult rat with unilateral fimbria fornix transection. Methods On the 2nd day after the right fimbria fornix of SD rats were transected, 5-bromodeoxyuridine (BrdU) was injected intraperitoneally once a day for 5 days. On the 28th day after operation, the brains were fixed for frozen section. BrdU/NF-200 double-label immunofluorescence was used to detect the proliferation and differentiation of NSCs into neurons. Results BrdU positive cells in the transected side of hippocampus were more than those in the normal side of hippocampus. The BrdU/NF-200 double labeled neurons were observed in the transected side of hippocampus, but absent in the normal side of hippocampus. Conclusions Fimbria fornix transection could cause endogenous NSCs in the hippocampus to proliferate rapidly and differentiate into neurons.
