摘要
目的探讨P-选择素在大鼠移植胰腺缺血再灌注损伤中的作用及P-选择素单克隆抗体的保护作用。方法75只SD大鼠随机被分为假手术组、移植组和治疗组。移植组和治疗组均进行全胰十二指肠移植术,但治疗组于再灌注前5min静注P-选择素单克隆抗体。3组分别于再灌注后1、3、6 h(n=5)取血测定血清淀粉酶水平,并切取胰腺标本进行组织病理学观察及P-selectin免疫组化染色。结果移植组胰腺组织损伤随再灌注时间的延长而加重,血淀粉酶升高,与中性粒细胞浸润直接相关;而治疗组胰腺组织损伤不明显,血淀粉酶降低。移植组各时段P-选择素均有阳性表达,且在再灌注1h达高峰,假手术组、治疗组P-选择素不表达。结论P-选择素在大鼠移植胰腺缺血再灌注损伤的病理过程中起重要作用。抗P-选择素单克隆抗体对移植胰腺缺血再灌注损伤有保护作用。
Objective To investigate the role of P-selectin (Ps) and the protection effect of monoclone antibodies against P-selectin on ischemia/reperfusion injury of rat pancreas graft. Methods Seventy five Sprague-Dawley rats were randomized into the transplanted group,therapeutic group and sham operation group.Both the transplanted group and the therapeutic group rats received whole pancreatico-duodenal transplantation.In the therapeutic group monoclone antibodies (mAb) against P-selectin were applied before reperfusion.Pancreas and blood samples from rats in the three groups were harvested at the 1st, 3rd and 6th(all n=5) after reperfusion for immunohistochemistry analysis of P-selectin, histopathology and serum amylase assay. Results ,(1)The damage of pancreas graft was aggravated gradually and the level of serum amylase was elevated markedly in the transplanted group as reperfusion was prolonged and was related to neutrophil infiltration,while these changes were mild in the mAb treated group.(2)Ps were expressed in the transplanted group and peaked at the first hour after reperfusion, but were not expressed in the sham operation and the therapeutic group. Conclusion (1)Ps plays important roles in ischemia/reperfusion injury of rat pancreas graft.(2)Anti-Ps mAb has protection effect on ischemia/reperfusion injury of rat pancreas graft.
