幼年特发性关节炎患儿趋化因子CCL5的检测及其意义

摘要 目的探讨趋化因子CCL5在幼年特发性关节炎(JIA)发病机制中的作用。方法采用定量PCR、流式细胞术及胞内染色的方法检测外周血及关节液单个核细胞CCL5的表达。结果JIA患者关节液单个核细胞高表达CCL5,而外周血中仅有少量表达,正常人外周血单个核细胞中未见CCL5的表达。CCL5主要表达于T细胞(CD3+CCL5+)。活化T细胞高表达CCL5,尤其以关节液中的T细胞表达水平增加更为明显。结论局部T细胞异常表达趋化因子CCL5可能参与了幼年特发性关节炎的发生。 Objective To explore the role of chemokine CCL5 in the patients with juvenile idiopathic arthritis (JIA). Methods CCL5 in peripheral mononuclear and synovial fluid was examined by using real-time PCR and intracellular flow cytomer. Results There was highly increased CCL5 level in the synovial fluid, while low level in peripheral blood. No CCL5 was detected in the normal controls. CCL5 was mainly expressed in T cells and activiated T cells expressed increased CCL5 compared with un-activiated T cells. Conclusion Abnormal highly expressed CCL5 may contribute to the local inflammation of JIA.
