摘要
目的观察乏氧诱导因子-1α(HIF-1α)和微血管密度(MVD)在S180肉瘤中的表达水平,并探讨两者间的相关性。方法建立昆明种小鼠S180肉瘤模型23个。待肿瘤生长至1~2cm时完整切取肿瘤组织,用免疫组化技术分别测定肿瘤组织的HIF-1α和MVD的表达情况。结果所有S180肉瘤组织的HIF-1α表达为7%~70%(中位数为21%),而MVD的表达为3~40条(中位数为10条);HIF-1α与MVD的表达成线性正相关(t=7.048,P<0.001)。结论S180肉瘤组织中均有HIF-1α和MVD不同程度的表达,并且两者间存在显著的正相关性。进一步证实了HIF-1α在高表达的情况下通过促进血管内皮生长因子的高表达而刺激肿瘤血管的生长。
Objective To investigate the expression of hypoxia inducible factor- 1 α ( HIF- 1α) and microvessels density (MVD), and further to evaluate the correlation between them. Methods Twenty- three Kunming mice bearing S180 sarcoma were set up. When S180 sarcoma grew to the size at between 1 and 2 cm, tumor was resected and the expression of HIF-1α and MVD was inspected by immunohistochemical method. Results The expression of HIF-1α ranged from 7% to 70% (median: 21% ) and the expression of MVD ranged from 3 to 40 strips(median: 10 strips). The further analysis found that the expression of HIF-1α and MVD had positive liner correlation( t = 7.048, P 〈 0. 001 ). Conclusion The results show that S180 sarcoma have the expression of HIF-1α and MVD in varying degree and there exists the positive liner relationship between them. It further proves that high expression of HIF-1α can stimulate vessel growth of tumor by regulation of high expression of vascular endothelial growth factor.
