摘要
目的利用大鼠肝脏热缺血再灌注(I/R)损伤模型,研究褪黑素(MT)对再灌注损伤肝脏的保护作用。方法用无损伤血管夹阻断部分肝脏血流,建立大鼠肝热缺血再灌注损伤模型;用缺口末端标记法(TUNEL法)检测肝细胞凋亡指数(AI);心脏采血,测定血浆ALT和AST的变化。结果模型组肝细胞AI升高,血浆ALT和AST升高(P<0.05),用褪黑素干预组肝细胞AI和血浆ALT和AST较模型组下降(P<0.05)。结论褪黑素可以抑制缺血再灌注损伤后肝细胞的凋亡,减轻肝脏的损伤。
Objective To study whether the melatonin(MT) can attenuate hepatocellular injury caused by ischemia-reperfusion(I/R) injury in rat liver. Methods I/R models were etablished by obstructing partial blood srteam with non-damage vascular clamp. The apoptosis of hepatocyte was detected by terminal deoxynucleotidyl transferase mediated dUDP-biotin nick end labeling(TUNEL). The serous ALT and AST were also detected. Results Compared with shame operated group, apoptosis index(AI) of hepatocyte increased significantly in I/R group( P 〈 0. 05), together with increase of serous ALT and AST( P 〈 0.05). Compared with the I / R group, the AI of hepatocyte decreased obviously in MT group, associated with decrease of serous ALT and AST( P 〈 0.05). Conclusion MT can inhibiting hepatocellular apoptosis and relieve the injury caused by I/R injury in rat liver.
