更年春方对卵巢切除大鼠学习记忆能力和海马ER亚型的调节 被引量:8

Regulative Effects of "Gengnianchun Decoction" on Learning and Memory and Hippocampal ER Subtype in Ovariectomized Rats

摘要 目的观察更年春方(GNC)对卵巢切除大鼠学习和记忆能力下降的预防和治疗作用,以及GNC对大鼠海马中雌激素核受体(ER)亚型的调节。方法采用10~12月龄SD雌性大鼠切除双侧卵巢建立学习和记忆能力下降的模型,用GNC灌药12周后行Morris水迷宫实验(MWM),实时荧光定量PCR法测定海马ERα和ERβmRNA的变化,免疫组化的方法测定海马CA3区ERα和ERβ蛋白的表达。结果大鼠切除卵巢后,血清E2和MWM成绩下降,海马中ERβmRNA与蛋白的表达均降低,而ERαmRNA和蛋白变化不明显。使用GNC者E2无明显变化,MWM成绩和海马中ERβmRNA与蛋白的表达提高,对ERαmRNA和蛋白无明显影响。结论提示GNC可能通过调节海马中的ERβ改善学习和记忆能力低下。 Objective To observe the effect of "C, engnianchun Decoction" (GNC) on learning and memory and ER subtype of the hippocampus in ovariectomized rats. Methods Ten to twelve months old SD rats were randomized into sham group, model group, high-dose GNC group and low-dose GNC group. These rats were fed with the Decoction or normal saline for 12 weeks respectively. The changes of leaxning and memory were tested by Morris water maze. On the second day of Morris water maze, blood was taken from the carotid artery of rats. The real time PCR was performed to determine the mRNA level of ERα and ERβ mRNA in right hlppocampuses. And the immunohistochemistry was performed to detect the distribution of protein of ERα and ERβ mRNA in paraffin section of left hippocampus. Results Twelve weeks after ovariectomy, serum estrogen level and learning-and-memory in Morris water maze was markedly decreased. And the scale of ERβ mRNA and protein decreased in ovariectomized rats, but the scale of ERα mRNA and protein was the same as sham group. The rats fed with the Decoction had better learning and memory and higher scale of ERβ mRNA and protein. Condusion It is suggested that "Gengnianchun Decoction" may be rather effective to improve learning and memory by regulating ERβ.
