盐酸甲氯芬酯对血管性痴呆大鼠学习记忆及ChAT表达的影响

摘要 目的探讨盐酸甲氯芬酯对血管性痴呆(VD)大鼠学习记忆的影响及可能机制。方法采用改良Pulsinelli’s四血管阻断法建立血管性痴呆大鼠模型;给予盐酸甲氯芬酯治疗后,用Mor-ris水迷宫检测学习记忆能力及免疫组化观测海马ChAT表达的变化。结果经盐酸甲氯芬酯治疗后,VD大鼠学习记忆能力提高,海马齿状回ChAT表达明显增加。结论盐酸甲氯芬酯改善VD大鼠学习记忆能力,可能与它保护神经细胞、增强ChAT表达有关。 Objective To investigate the effects of centrophenoxine on the learning and memory of the vascular dementia (VD) model rats and its possible mechanisms, Methods The VD animal model was established by employing improved method of Pulsinelli's four -vessel occlusion, and then the model rats were treated with centrophenoxine. Morris maze testing was used to evaluate the learning and memory of the rats, and the expression of choline acetyltransfrase (CHAT) in the hippocampus was investigated by using immunohistochemistry. Results After treatment of the VD rats with centrophenoxine, the learning and memory was improved significantly, and the expression of CHAT in the dentate gyrus of the hippocampus increased. Conclusion Centrophenoxine enhances the learning and memory of the model rats, that possibly it protecting neurons and increasing the expression of CHAT in the hippocampus.
