原癌基因c-raf在精原干细胞中的表达及作用

The Expression and Effect of Proto-oncogene c-raf in Spermatogonial Stem Cells

摘要 本研究用基因测序和BLAST检验扩增的精原干细胞DNA和c-raf的同源性;用MTT法观察c-rafASODNs(反义c-raf寡脱氧核甘酸)拮抗c-raf对体外培养精原干细胞培养的作用,探讨原癌基因c-raf对体外精原干细胞培养增殖的影响.实验结果显示扩增的精原干细胞DNA和c-raf的同源性98%;c-rafASODNs对体外培养的精原干细胞存活呈剂量依赖性抑制作用(P<0.05).提示c-raf在精原干细胞中表达;原癌基因c-raf促进精原干细胞体外培养的增殖. DNA sequencing and BLAST were used to identify the homology between the amplified spermatogonial stem cell's DNA and c-raf. The effect of antisense oligodeoxynucleotides(ASODNs) on proliferation of spermatogonial stem cells in vitro was observed by MTT assay. The results showed that the homology between the amplified spermatogonial stem cell's DNA and the c-raf was 98 % and c-raf ASODNs decreased the survival rate of spermatogonial stem cells in a does-dependent manner (P 〈 0.05). It's suggested that c-raf was not only expressed in spermatogonial stem cell but also enhanced the survival and proliferation of spermatogonial stem cell.
