摘要
目的:探讨脂多糖(Lipopolysaccharide,LPS)诱导细胞滋养细胞凋亡的机制。方法:从正常早孕绒毛分离细胞滋养细胞,用无血清培养基培养。在培养基中加入不同浓度的LPS,使其终浓度分别为0、25、50、100、200ng/ml。用光镜和透射电镜观察细胞滋养细胞的形态学变化;用Western blot检测Bax、Bcl-2和Caspase-8的表达。结果:LPS作用后24h,光镜和电镜下见细胞滋养细胞出现凋亡的形态学变化;Western blot检测结果表明,LPS抑制Bcl-2表达,促进Bax和Caspase-8表达。结论:LPS能够经线粒体途径诱导细胞滋养细胞凋亡。
Objective:To investigate whether Lipopolysaccharide(LPS) can induce apoptosis in cytotrophoblasts via mitochondria pathway. Methods:Villous were collected from normal early pregnant abortions. Then cytotrophoblasts were isolated and cultured with serum-free media. Subsequently, cytotrophoblasts were treated with LPS at the concentrations of 0 (control) ,25,50,100 and 200ng/ml. Apoptosis in cytotrophoblast was tested by light microscope, and transmission electron microscope. The expressions of Bcl-2, Bax, and Caspase-3 were detected by Western blot. Results:Under light and transmission electron microscope, characteristic pathological changes of apoptosis in cytotrophoblast were observed after LPS treatment. Western blot results showed that LPS inhibited expression of Bcl-2 and promoted expression of Bax and Caspase-8. Conclusion:LPS induces apoptosis in cytotrophoblasts via mitochondria pathway.
