呼吸道合胞病毒蛋白G基因核苷酸分析

摘要 目的:对河南省呼吸道合胞病毒(RSV)进行分离与鉴定。方法:选择2006年冬季至2007年春季河南省部分医院急性呼吸道感染患者,采集咽拭子,利用Hep-2细胞和Vero细胞进行病毒分离,对分离株用特异性RT-PCR鉴定;对鉴定阳性的RSV毒株进行蛋白G全基因序列测定,并进行同源性分析,构建基因进化树。结果:共分离到4株RSV毒株,各分离株之间核苷酸同源性92%~99%;与A型RSV标准参考株的核苷酸同源性为86%~92%,与B型RSV标准株的同源性为70%。结论:2006年河南省RSV流行株为A型。 Aim: To isolate and identify respiratory syncytial virus(RSV) from Henan province. Method: RSV was isolated from swab collected from patients with syndrome of fever, cough in some Henan hospitals in the winter of 2006. The isolated strains identified by RT-PCR, were subjected to sequence the whole region of the G gene, and phylogenetic trees were constructed by comparison with RSV standard strains and reference strains. Results: Four RSV strains were isolated. The identities of the G'gene among the four isolated strains were 92% -99% ,the identity of the G gene between the isolated strains and the reference A strain was 86% -92% ,but with the B strain, the identity was 70%. Conlusion:RSV prevalent in Henan province in the winter of 2006 is type A.
