巴戟天醇提物对血淤大鼠血流动力学及血小板聚集的影响 被引量:10

摘要 目的:观察巴戟天醇提物对血淤大鼠血流动力学及血小板聚集率的影响。方法:60只Wistar大鼠随机等分为正常对照组、模型组、复方丹参滴丸(0.1g/(kg.d))阳性药对照组及巴戟天醇提物12g/kg、6g/kg、3g/kg剂量组6组。正常对照组和模型组给予等体积的5g/L羧甲基纤维素钠溶液,其他各组每d1次灌胃给药,连续5d。采用2次皮下注射肾上腺素1mg/kg之间将大鼠置于冰水中5mim的方法建立大鼠急性"血淤"模型,观察药物对血淤模型大鼠高、中、低切变率下全血黏度、全血卡森黏度、血浆黏度、红细胞压积、全血还原黏度、红细胞电泳时间及二磷酸腺苷、胶原、凝血酶诱导血小板最大聚集率的影响。结果:巴戟天醇提物3个剂量组均能降低血淤大鼠的高、中、低切变率下全血黏度、全血卡森黏度,缩短红细胞电泳时间,抑制血小板最大聚集率(P均<0.01);其高、中剂量组尚能降低全血还原黏度(P<0.01或0.05)。结论:巴戟天醇提物可改善血淤时的血流动力学,抑制血小板聚集。 Aim: To investigate the influence of Radix morindae officinalls(RMO) ethanolic extracts on the hemorheology and platelet aggregation in acute stress blood stasis rats, Methods:A total of 60 Wistar rats were randomly separated into six groups ( 10 rats per group) :normal control group, model group, Fu Fang Dan Sen Di Wan (0.1 g/kg) group, RMO ethanolie extracts 12 g/kg,6 g/kg,3 g/kg groups, Acute stress blood stasis rats model was prepared by leaving rats in ice water for 5 minutes between two times cutaneously injecting of hyodermic epinephrine 1 mg/kg,and the main hemorheological indexes and the rate of platelet aggregation induced by ADP, COL,and thrombin were detected, Three different concentrations ( 12 g/kg,6 g/kg,3 g/kg) of RMO ethanolic extracts was given daily for five days via oral administration, Results: 12 g/kg, 6 g/kg,3 g/kg treatment of RMO not only significantly reduced the increase of whole blood viscosity at either high, middle, or low shear rates, whole blood easson viscosity and red-cell electrophoresis time,but also restrained the rate of platelet aggregation (P 〈 0.01 ), In addition, 12 g/kg,6 g/kg treatment of RMO ethanolic extracts also decreased the whole blood reduction viscosity in acute stress blood stasis rats (P 〈 0. 01 or 0.05), Conclusion : RMO ethanolie extracts has the ability of blood-activating and tasis-eliminating.
