In vivo experiment study of non-ablative photorejuvenation using Q-Switched Nd:YAG laser in rats

摘要 目的:评估Q开关Nd:YAG激光非损伤嫩肤后小鼠真皮胶原蛋白含量的变化,为临床治疗提供理论依据。方法:采用532nm,1064nm的Q开关Nd:YAG激光;对32只小鼠随机分为4组,1组对照组,3组实验组。分别用532nm、1064nm以及这两种波长激光联合各照射5次,间隔7天。免疫组化技术检测32只小鼠真皮中Ⅰ和Ⅲ型胶原蛋白含量,HE染色测定真皮胶原纤维的厚度。结果:(1)胶原纤维的厚度及Ⅰ和Ⅲ型胶原蛋白的含量均比对照组高(P<0.01);(2)532nm组的胶原纤维的厚度及Ⅰ和Ⅲ型胶原蛋白的含量均比1064nm组高(P<0.01);两种波长联合的胶原纤维的厚度及Ⅰ和Ⅲ型胶原蛋白的含量均比532nm组和1064nm组高(P<0.01)。结论:532nm、1064nm以及这两种波长的激光联合照射是有效的嫩肤方法,532nm优于1064nm,两种波长联合优于单用一种波长。 Objective: To evaluate the change of the amount of collagen in the dermis after Q - Switched Nd: YAG laser non - ablative photorejuveantion, in order to offer reference for effective clinical treatment, nethods: Thirty- two rats were randomly divided into four groups including three laser irradiation groups and one control group. The rats' skin was irradiated by 532 nm, 1064 nm and a combination of 532 nm and 1064 nm Q- Switched Nd:YAG laser for five times at interval of seven days. The content of collagen type Ⅰand Ⅲ in the dermis was measured by immunohistochemical method. The thickness of collagen bundles in the dermis was measured under HE stain. Results: (1) The collagen fiber was thicker and the content of collagen type Ⅰand Ⅲ was higher in the experimental groups than in the control group ( P 〈 0.01 ), in the 532 nm group than in the 1064 nm group ( P 〈 0.01 ), and in the combination group of 532 nm and 1064 nm than in 532 nm alone and in 1064 nm alone ( P 〈 0.01 ). Conclusion: A combined irradiation of 532 nm and 1064 nm yield better result than either used alone, and 532 nm irradiation is superior to 1064 nm.
