针刺足三里调控胃肠动力机制的实验研究 被引量:27

The mechanism of electroacupuncture for regulating gastrointestinal motility in rabbits

摘要 目的探讨针刺足三里调控家兔胃肠动力的机制。方法选择健康成年实验用家兔30只,分为对照组、针刺足三里穴组及针刺非穴位组。在针刺前后多点(针刺前及针刺后15、30、45、60 min)采血测定胃动素含量,并在针剌1h后处死动物,取胃和空肠组织进行胆碱酯酶染色、一氧化氮合酶染色并电镜观察神经末梢囊泡。结果针剌足三里穴后家兔血浆胃动素含量增多,并在针刺后45min达高峰。胃及近端空肠平滑肌组织内的胆碱酯酶活性在针刺足三里穴组明显增高。与对照组比较差异有统计学意义[胃:(15571±2876)像素点比(9081±801)像素点,近端空肠:(18588±4909)像素点比(11703±2050)像素点.P<0.01]。一氧化氮合酶活性在针剌足三里穴组明显降低,与正常组比较差异有统计学意义(胃:1.38±0.74比4.71±1.06,近端空肠:2.10±0.94比4.81±1.08,P<0.01)。针刺足三里穴后,胃肠道平滑肌组织神经末梢内含乙酰胆碱递质的无颗粒囊泡数目明显减少,与正常对照组比较差异有统计学意义(胃:31.12±9.99比57.45±9.18,近端空肠:21.89±4.35比56.70±9.35,P<0.05)。结论胃动素、胆碱能神经及非肾上腺素能非胆碱能神经参与了针剌足三里穴对胃肠道运动的调节机制。 Objective To investigate the mechanism of regulating gastrointestinal motility by electroacupunturing point of "Zusanli" in rabbits. Methods Thirty adult rabbits were divided into control, electrical acupuncture point of "Zusanli" and non acupuncture point groups. The blood concentrations of motilin were detected at different times(before acupuncture and 15, 30, 45 and 60 min after acupuncture). Sixty rain after acupuncture, the mice were sacrificed . The acetylcholine esterase and nitric oxide synthase(NOS) in gastric and jejunum tissues were examined. The electron microscope was used to observe the vesicles of nerve ending. Results The concentrations of motilin in electrical acupuncture group was significantly increased and reached the high peak at 45 min. The activities of acetylcholine esterase was increased significantly in electrical acupuncture group compared to control group [gastric tissue: (15 571 ± 2876)pinel vs (9081± 801)pinel, P〈 0.01) and jejunum tissue: (18 588 ± 4909)pinel vs (11 703 ± 2050) pinel, P 〈 0.01]. The activities of NOS in electroacupuncture group were significantly decreased compared to control group (gastric tissue: 1.38 ± 0.74 vs 4.71 ± 1.06 and jejunum tissue: 2.10 ± 0.94 vs 4.81 ± 1.08, P 〈 0.01). The electron microscopy revealed that the number of vesicle without granula was significantly decreased in electrical acupuncture group compared to control group (gastric tissue: 31.12±9.99 vs 57.45 ± 9. 18 and jejunum tissue: 21. 89 ± 4. 35 vs 56. 70 ± 9. 35, P 〈 0.05). Conclusion Motilin, cholinergic nerve and non adrenergic/non-cholinegic nerve may be involved in the mechanism of regulating gastrointestinal motility by acupuncturing pointy of "Zusanli'.
