Glypican-3基因在肝细胞癌组织中的表达

摘要 目的观察Glypican-3基因在肝细胞癌中的表达和突变关系。方法采用RT-PCR和SSCP分别检测48例肝细胞癌肿瘤组织、癌旁组织和正常肝组织中Glypican-3 mRNA的表达和基因突变。结果Glypican-3 mRNA在肿瘤组织中的阳性表达率为77.1%,癌旁和正常肝组织中未见表达;Glypican-3表达与肝癌临床分期、细胞分化程度均有显著相关性(P<0.05);肝细胞癌组织中未发现Glypican-3基因突变。结论正常基因型Glypican-3在肝细胞癌中的过度表达可能是作为"安全卫士"来对抗癌基因的异常表达,从而发挥抑癌作用。 Objective To investigate Glypican-3 gene expression and mutation in hepatocellular carcinoma (HCC).Methods Glypican-3 gene expression and mutation in tumor,para-eancer and normal tissue of 48 HCCs were detected by RT-PCR and single-strand conformation polymorphism(SSCP), respectively. Results There was no Glypican-3 mRNA expression in para-cancer and normal tissue. Expression rate of Glypican-3 mRNA was 77. 1 % in tumor tissue, which was correlated with clinical staging and cell differentiation(P 〈 0,05), No mutation of Glypican-3 gene occurred in HCC.Conclusion Overexpression of normal genotype Glypican-3 in HCC might serve as a "security bodyguard", which antagonized the abnormal expression of oncogenes, and thus played an inhibitory role in the carcinogenesis.
