摘要
目的探讨肾脏间质树突状细胞在多器官功能障碍综合征免疫紊乱中的影响与作用。方法腹腔注射酵母多糖制造C57BL/6小鼠MODS模型,分为正常、3~6 h、12~48 h、5~7d和10~12 d组。观察各组小鼠肾脏间质树突状细胞的病理学变化及其表面标记物CD11c、CIY205、CD80和I-A^b在MODS各期肾脏中的表达水平;流式细胞术检测外周血CD4^+与CD8^+的T细胞数量与比值。结果在急性损伤期,肾脏间质树突状细胞大量增生,CD 80和I-A^b表达较正常组差异具有统计学意义(P<0.01和P<0.05),外周血T细胞CD^4/CD8^+比值则明显下降(P<0.01)。功能衰竭期,肾脏间质树突状细胞增生至最高点,但其CD80和I-A^b表达较急性损伤期显著减少(P<0.01),外周血T细胞CD4^+/CD8^+比值下降至最低(P<0.01)。结论肾脏间质树突状细胞参与并影响了MODS中肾脏局部与全身的免疫失衡及免疫抑制过程。
Objective To explore the role of kidney interstitial dendritic cells in immunodissonance moechanism in mice with multiple organ dysfunction syndrome ( MODS). Method The model of MODS wasmade by injecting zymosan into the peritoneal caiecty of C57BL/6 mice, and the mice were randomly divided into 5 groups, namely, normal, 3 - 6 hours, 12 - 48 hours, 5 - 7 days, 10 - 12 days after administrating zymosan. Pathological changes of kidney interstitial dendritic cells were observed by transmission electronic microscopes. Specific surface markers CD205, CD11c, CD80, MHCⅡ molecules I-A^b, CD4^+ and CD8^+ T lymphocyte subgroups in peripheral blood were detected by immunohistochemistry and flow cytomctry. Results In acute injury stage, in comparison with normal group, interstitial dendritic cells had a continuous proliferation with high expression of CD80 and I-A^b ( P 〈 0.01 and 0.05). The ratio of CD4^+/CD8^+ declined markedly in peripheral blood (P 〈 0,01 ). In renal failure stage, interstitial dendritic cells further proliferated to the maximum magnitude, but the expression of CD80 and I-A^b declined to very low level ( P 〈 0.01 ), compared with that in acute injury stage and the ratio of CD4^+/CD8^+ declined remarkably to the minimum in periphend blood ( P 〈 0.01). Conclusions Renal interstitial dendritic cells participate in and influence the course of immunological dysfunction and immunodepression of local kidney and systemically in MODS.
