肝移植术后外周血单个核细胞与CD34^+细胞中HBV DNA含量及复制状态检测 被引量:3

摘要 PBMC包括T细胞、B细胞和单核细胞等,均由造血干细胞分化而来。HBV相关肝病肝移植术后,受体PBMC内仍有HBV DNA存在,是HBV再感染的主要病毒来源。在肝移植术后持续规范的拉米夫定联合乙型肝炎免疫球蛋白治疗及细胞半衰期作用下,PBMC应逐渐呈HBV DNA阴性,但临床情况并非如此,推测造血干细胞已有HBV感染,其内的HBV随细胞分裂而复制并进入子代细胞,导致HBV持续存在及耐受。为证实此推测,我们运用密度梯度离心结合磁珠分选,分离PBMC中的T细胞、B细胞、单核细胞和骨髓CD34^+细胞并提取DNA,进行总HBV DNA和HBV共价闭合环状DNA(cccDNA)实时荧光PCR定量检测。
