液相色谱-电喷雾离子阱质谱联用确证小鼠灌胃给药CTN986后血与脑组织中的药物原型 被引量:1

摘要 目的:确定 CTN986小鼠灌胃给药后血与中枢神经系统(CNS)中的药物原型。方法:血浆、脑组织样品用甲醇沉淀蛋白,离心后取上清液经固相萃取后用液相色谱-离子阱质谱进行测定。结果:小鼠灌胃给药后的血和脑组织样品经液质测定,所得的选择离子流色谱图(m/z 741)及一、二、三级质谱图均与对照品 CTN986选择离子流色谱图、各级质谱图一致。结论:小鼠灌胃给药后,CTN986能以原型的形式吸收入血并且可以通过血脑屏障进入中枢。 Objective : To identify parent drug of CTN986 in mice blood and brain after ig administration. Methods: Samples of plasma and brain homogenate were added methanol to precipitate proteins and supernatants were identified by high performance liquid chromatography - electrospray ion trap mass spectrometer after solid phase extraction. Results: The SIM (m/z 741 )chromatograph and bar graphs of MS1 , MS2 and MS3 of plasma and brain homogenate samples conform to their respective standard. Conclusion:After mice were administered via ig, CTN986 was absorbed into blood and distributed to brain through the blood - brain barrier in the form of parent drug.
