HPLC法测定鸡骨草药材中相思子碱的含量

HPLC determination of abrine in Abrus cantoniensis Hance

目的:建立测定鸡骨草药材中相思子碱含量的高效液相色谱法。方法:Kromasil C_(18)色谱柱(250 mm×4.6 mm,5 μm),流动相为甲醇-水-磷酸-三乙胺(12:88:0.36:0.16),流速1.0 mL·min^(-1),检测波长280 nm。结果:对不同来源的17批鸡骨草药材进行了测定,其相思子碱的含量在0.02%～0.66%之间。相思子碱在0.1～4.0μg范围内线性关系良好(r=0.9999,n=8),平均回收率为99.2%(n=9)。结论:该方法可以用于鸡骨草药材的质量控制。

Objective： To establish a method for determination of abrine in Abrus cantoniensis Hance by HPLC. Methods： The separation was performed in a Kromasil C18 column （250 mm×4. 6 mm, 5 μm）with a mobile phase of methanol - water - phosphoric acid - triethylamine （ 12：88： 0. 36： 0. 16）. The flow rate was 1.0 mL·min^-1, and the detecting wavelength was at 280 nm. Resdts： 17 batches of the herb of A. cantoniensis from different habitats were detected and the contents of abrine ranged from 0.02% to 0. 66%. The linear range was 0. 1 -4. 0 μg（ r = 0. 9999,n =8）. The mean recovery of abrine was 99.2% （n = 9）. Condusion： The method can be used in the quality control of A. cantoniensis.
