Lox-1基因501G>C和3′UTR*188C>T多态性与急性心肌梗死发病的相关性 被引量:4

Lack of Association of 501G>C, 3′-UTR*188C>T Polymorphisms of the Oxidized LDL-Receptor 1 (OLR1) Gene in Patients with Acute Myocardial Infarction

摘要 目的检测Lox-1501G>C和3′UTR*188C>T多态性在中国汉族人群中的分布及其与急性心肌梗死(acute myocardial infarction,AMI)发病的相关性。方法利用荧光染料SYBRGreenⅠ标记定量PCR产物,通过PCR生长曲线和融解曲线分析结果进行多态位点的基因分型。分别检测汉族202名AMI患者及161名健康人群Lox-1501G>C和3′UTR*188C>T多态性。随机选取30份经荧光定量PCR分型的标本进行DNA测序鉴定。结果AMI患者和健康人群的Lox-1501G等位基因频率分别为0.834和0.798,Lox-1501C等位基因频率分别为0.166和0.202,Lox-1501G>C基因型频率在AMI组和对照组间差异无统计学意义[OR0.687,95%CI(0.226~2.093);P=0.507]。AMI患者和健康人群的Lox-13′UTR*188C等位基因频率分别为0.681和0.674,Lox-13′UTR*188T等位基因频率分别为0.319和0.326,Lox-13′UTR*188C>T基因型频率在AMI组和对照组亦无统计学意义[OR0.947,95%CI(0.623~1.439);P=0.80]。结论中国汉族Lox-1基因501G>C和3′UTR*188C>T多态性与AMI发病无关。 Objective To investigate 501G〉C and 3' UTR^* 188C〉T polymorphisms of Lox-1 in Chinese Han population and the association of these polymorphisms with acute myocardial infarction (AMI). Methods The quantitative fluorescence PCR approach was used to determine the 501G〉C and 3' UTR^* 188C〉T polymorphisms of Lox-1 gene based on PCR growth curves and melt curve analysis with the indicating fluorophore SYBR Green I. The two polymorphisms of Lox-1 gene were determined by quantitative PCR approach in 202 AMI patients and 161 unrelated healthy controls. The accuracy of genotyping of 30 samples determined by quantitative fluorescence PCR was validated by DNA sequencing. Results The results of Lox-1 genotyping determined by quantitative fluorescence PCR were consistent with the results of DNA sequencing. The frequency of Lox-1 501G allele was detected in 0.834 of AMI patients and 0.798 of healthy controls, and Lox-1 501C allele was detected in 0.166 of AM I patients and 0.202 of healthy controls. Individuals possessing a Lox-1 501^*C genotype do not have a higher risk ofAMI IOR 0.687, 95% CI (0.226-.093) ;P=0.507]. The frequency of Lox-1 3' UTR^*188C allele was detected in 0.681 of AMI patients and 0.674 of normal controls, and Lox-1 3' UTR^*188T allele was detected in 0.319 of AMI patients and 0.326 of healthy controls. Individuals possessing a Lox-1 3' UTR^* 188 *T genotype also do not have a higher risk ofAMI IOR 0.947, 95% CI (0.623-1.439) ;P=-0.80]. Conclusion The Lox-1 genotyping results suggested that Lox-1 501G〉C and 3' UTR^* 188C〉T polymorphisms are not associated with AMI in Chinese Han population.
