流式细胞术分析T细胞亚群—两种荧光染色方法的比较

T LYMPHOCYTE SUBSETS ANALYSIS BY FLOW CYTOMETRY—COMPARISON OF TWO DIFFERENT FLUORESCENCE STAINING METHODS

应用鼠抗人Ｔ细胞单克隆抗体ＯＫＴ系列，结合流式细胞术，分别用直接免疫荧光法（ＤＦＡ）和间接免疫荧光法（ＩＦＡ）对正常成人外周血Ｔ细胞亚群进行了检测。结果表明，两种方法测定的ＣＤ＋３、ＣＤ＋４和ＣＤ＋８细胞百分率以及ＣＤ＋４／ＣＤ＋８比值均无显著性差异（Ｐ＞０．０５），但ＤＦＡ检测的非特异性荧光染色细胞数（２．８１±２．０２）明显低于ＩＦＡ（１１．５７±５．５９）。

Two methods of T lymphocyte subsets determination in the peripheral blood of normal adults by OKT monoclonal antibody and flow cytometry are reported.They are direct immunofluorescence assay (DFA)and indirect immunofluorescence assay(IFA).Our studies showed that the results obtained by both methods (the percentage of CD 3 +,CD 4 + and CD 8 + cells and the ratio of CD 4 +/CD 8 +)were in good agreement with each other(p>0.5). But the cell numbers of non specific fluorescent stainning by DFA was lower than those by IFA,and each method had its own experimental specialities.
