摘要
目的 了解印迹基因泛素蛋白酶基因(UBE3A)在人类卵母细胞和植入前胚胎的表达状况。方法采用单细胞巢式RT-PCR,检测UBE3A在人类卵母细胞和植入前各个阶段胚胎中的mRNA表达。结果UBE3A在人类卵母细胞MⅠ、MⅡ和植入前胚胎2、4、6、8细胞期均有表达,单卵母细胞扩增成功率为83.3%,单个卵裂球细胞为72.3%,平均扩增成功率为77.9%。单个卵母细胞和单个卵裂球扩增成功率无显著性差别(P〉0.05),但单卵母细胞较单个卵裂球高。结论UBE3A在人类卵细胞和植入前胚胎均有表达,有助于研究卵母细胞成熟及胚胎生长发育,为探讨印迹基因与辅助生殖及遗传性印迹疾病的关系提供理论依据。
Objective To detect the expression of imprinted gene UBE3A in human oocytes and preimplantation embryos. Methods The expression condition of imprinted gene UBE3A in human oocytes and pre-implantation embryos was detected by using of the monoplast nested reverse transcription and polymerase chain reaction amplification (RT-PCR). Results UBE3A was expressed in the metaphase Ⅰ( M Ⅰ ) and metaphase Ⅱ ( M Ⅱ ) of human oocytes and in the 2-cell, 4-cell, 6-cell and 8-cell stages of pre-implantation embryos. The success rate of amplification in single oocyte and individual cleavage was 83.3% , and 72. 3% , respectively. The average success rate was 77.9%. The success rate of amplification was no statistical significance between single ooeyte and individual cleavage( P 〉 0. 05). Conclusion The expression of UBE3A gene existed in human oocytes and pre-implantation embryos, which conduces to study of oocyte maturation and embryo development. It provided the theoretical basis for investigating the correlation among imprinted genes, assisted reproductive technology and genetic imprinted diseases.
