摘要
【目的】通过观察谷氨酰胺对内毒素血症时幼年大鼠肾脏细胞凋亡超微结构的影响,探讨谷氨酰胺对内毒素血症大鼠肾脏损伤的保护机制。【方法】健康18日龄Wistar大鼠54只,随机分为3组:正常对照组(N):腹腔注射生理盐水0.1ml;内毒素组(L):腹腔注射内毒素4 mg/kg;谷氨酰胺干预组(G):腹腔注射内毒素同时腹腔注射N(2)-L-丙氨酰-L-谷氨酰胺2 g/kg,于注射后6 h2、4 h和72 h处死大鼠取出肾脏,利用透射电镜观察肾脏细胞超微结构的改变。【结果】注射LPS 6 h、24 h肾小球基底膜厚薄不均,局部有增厚和足突融合,近端小管上皮细胞内线粒体嵴溶解,远端小管微绒毛减少;注射LPS 72 h近端小管出现核染色质的浓缩、着边及核破碎的凋亡改变,应用谷氨酰胺后肾脏结构改变明显减轻,凋亡改变消失。【结论】细胞凋亡参与内毒素血症幼年大鼠肾损伤过程,谷氨酰胺可减轻肾脏细胞凋亡,保护肾脏功能。
【Objective】To explore the mechanism of endotoxemic injury in kidneys by observing the effect of glutamine on ultrastructure of renal cells in rats with endotoxemia.【Methods】Fifty four eighteen-day Wistar rats were divided into three groups randomly.Control group: animals were injected intraperitoneally with the same volume(0.1 ml) of 0.9% sodium chloride as other two groups.LPS group: animals were injected by a single bolus of lipopolysaccharide(LPS 4mg/kg).Treatment group: animals received LPS 4mg/kg + glutamine 2g/kg.Then they were sacrificed at 6h、24h and 72h after injection.The ultrastructure was observed by electron microscope.【Results】Glomerular basement membrane became thick and foot processes had coalescent partly,mitochondrial cristae became dissolved in proximal tubular endothelial cells and microvillus of distal tubules diminished at 6h、24h after LPS injection.The morphological changes of apoptosis were found in the proximal tubules at 72h after LPS injection.Then these changes were relieved when glutamine was used.【Conclusion】Apoptosis participates in endotoximic injury of kidneys and glutamine has protective effect on injuried renal cells.
