Survivin、MMP-2在肾透明细胞癌组织中的表达及关系

Expression of Survivin,MMP-2 in Human Renal Clear Cell Carcinoma

[目的]探讨Survivin、MMP-2在肾透明细胞癌（renal clear cell carcinoma,RCCC）中的表达及意义。[方法]应用免疫组化技术检测Survivin、MMP-2在55例RCCC和20例正常肾组织中的表达。[结果]从正常肾组织到RCCC组织,Survivin、MMP-2阳性表达量显著升高（P〈0.05）。Survivin的表达与肿瘤大小无明显关系（P〉0.05）,与病理分级、临床分期密切相关,G3-G4组、Ⅲ-Ⅳ期组中Survivin的表达明显高于G1-G2组、Ⅰ-Ⅱ期组。MMP-2以肿瘤大小、病理分级、临床分期做分析,两组阳性表达率无明显差异（P〉0.05）。有淋巴结转移患者的Survivin、MMP-2阳性表达量较其对应组增多（P〈0.05）。[结论]Survivin蛋白异常表达在肾透明细胞癌的恶化、侵袭、转移中起重要作用,与RCCC组织细胞生物学特点密切相关,MMP-2在RCCC组织中阳性表达增高。可能在RCCC发生,发展、组织侵袭、转移中起重要作用。Survivin和MMP-2可能成为RCCC的重要诊断指标和治疗靶点。

[Objective]To study the expression and significance of Survivin and MMP-2 in human renal clear cell carcinoma（RCCC）.[Methods]Immunohistochemical method was used to detect Survivin and MMP-2 in 55 cases of RCCC and 20 cases of normal kidney.[Results]The positive rate of Survivin expression in RCCC was 65.5%（36/55）,higher than in normal renal tissue（0%,0/20）（P〈0.01）.There was relationship between Survivin expression and clinical stage and tumor grade of differentiation,but no relationship between Survivin expression and tumor size.The positive rate of MMP-2 expression in RCCC was 61.8%（34/55）,higher than in normal renal tissue（25%,5/20）（P〈0.01）.There was no relationship between MMP-2 and tumor size,tumor differentiation grade,or clinical stage.There was relationship between Survivin,MMP-2 and lymphonode metastasis（P〈0.05）.[Conclusion]Survivin and MMP-2 may play important roles in occurrence and progression of RCCC.Over-expression of survivin and MMP-2 indicates poor prognosis.Survivin and MMP-2 can be important diagnostic/therapeutic targets in RCCC.
