摘要
【目的】探讨不同浓度的碱性成纤维细胞生长因子(BFGF)对体外培养肌腱细胞增殖和量效关系的影响,并初步估计促增长的最佳浓度。【方法】PV-9000二步法免疫组化细胞定性,MTT法染色,在酶联免疫检测仪上测出不同浓度BFGF组光密度(optical density,OD)均值。【结果】与对照组OD均值相比:1.0、2.0、5.0、10、30、40、50 ng/mL BFGF组有显著性差异(P〈0.05);0.5、20 ng/mL BFGF组无显著性差异(P〉0.05)。各组OD均值随BFGF浓度的增加,先逐渐增大(0~5.0 ng/mL),达到峰值后,又逐渐减少(5.0~50 ng/mL)。【结论】1.0、2.0、5.01、0 ng/mL BFGF有促进肌腱细胞增殖作用;0.5和20 ng/mL BFGF对肌腱细胞增殖无明显作用;30、405、0 ng/mL有抑制肌腱细胞增殖作用。5.0 ng/mL是促肌腱细胞增殖最佳BFGF浓度。
[Objective]To study effects of BFGF of different concentration on proliferation and dose-effect relationship of tendon cells cultured in vitro and estimate the optimal BFGF concentration to promote proliferation routinely. [Methods]The groups of different BFGF concentrations were stained by MTT methods and their optical density(OD) average values were detected by enzyme linked immunodetection meter. [Results]By contrast to OD mean of control group: 0.5,1.0,2.0,5.0 ,30,40 ,50ng/mL BFGF group had significant difference( P〈0.05) ;0.5 and 20ng/mL BFGF group had no significant difference ( P〉0.05). When BFGF concentration varying from 0ng/mL to 5.0 ng/mL, OD mean of every group increased gradually , while from 5.0 ng/mL to 50 ng/mL, it decreased step by step. [Conclusion]BFGF of could promote proliferation of tendon cells; 0.5ng/mL and 20ng/mL BFGF has no significant effect on proliferation of tendon cells; 30ng/mL, 40ng/mL and 50ng/mL BFGF could inhibit proliferation of tendon cells. And 5.0ng/mL is the optimal BFGF concentration to promote proliferation.
