补肾化瘀及补肾健脾化瘀调整小鼠免疫衰老的对比研究 被引量:19

摘要 根据中医理论、临床经验及实验研究,我们认为“正虚(主要指脾肾气虚)夹瘀是导致衰老的主要原因/机制。为了验证上述观点,探讨中医衰老的本质和延缓衰老的有效方法和途径,本课题通过体内外实验从淋巴细胞IL-2产生的水平、自由基代谢、细胞膜流动性等方面对比研究了补肾化瘀八味丹坤汤(补化)和补肾健脾化瘀八味参芪丹坤汤(补健化)的抗衰老作用。实验结果表明,八味参芪丹坤汤提高老年小鼠和臭氧早衰模型小鼠IL-2产生的水平优于八味丹坤汤(p<0.01,p<0.05)。体外实验也表明八味参芪丹坤汤对恢复老年小鼠和H_2O_2损伤的淋巴细胞产生IL-2的作用均优于八味丹坤汤(p<0.01,p<0.05)。八味参芪丹坤汤在提高老年小鼠红细胞自由基清除酶SOD(超氧化物歧化酶)和CAT(过氧化氢酶)活性方面也优于八味丹坤汤(p<0.05)。并且,八味参芪丹坤汤组(补健化组)与八味丹坤汤组(补化组)相比,其稳定老年小鼠淋巴细胞膜流动性的效果更佳(p<0.05)。综上结果可见,补肾健脾化瘀复方八味参芪丹坤汤在调整衰老小鼠免疫功能、改善自由基代谢、抗自由基损伤和改善细胞膜功能等方面均优于补肾化瘀八味丹坤汤。其结果为“正虚夹瘀是衰老的主要原因/机制”,以及“补肾健脾化瘀法是延缓衰老的主要途径”的论点提供了实验依据。 According to the theory of TCM, clinical experience and experimental study, we hold that Zhengxujiayu (deficiency of kidney and spleen complicated with blood stasis) is the main cause/ mechanism of aging. In order to demonstrate the concept mentioned above, approach the essence of aging in TCM and search for the best therapeutic method and route of delaying aging, we have made comparative study between the decoction of tonifying kidney invigoration spleen removing blood stasis (BSQDKT) and the decotion of tonifying kidney invigoration spleen removing blood stasis (BDKT)on IL-2 production of splenic lymphocytes, radicals metabolism and membrane fluidity of splenic lymphocytes in vivo and in vitro. The results showed that BSQDKT offered some advantages over BDKT in improving the IL-2 production in old and ozone inhaled aging model mice (p<0. 01, p<0. 05). Experiments in vitro also demonstrated that BSQDKT was more effective than BDKT on restoring IL-2 production of splenic lymphocytes from old mice and normal mice injured by H202 (p<0. 01, p<0. 05). The activities of SOD and CAT of erythrocytes in old mice with BSQDKT also showed a greater increase than that with BDKT (p<0. 05). Meanwhile, BSQDKT is also more effective on stabilizing cell membrane fluidity in old mice than BDKT (p< 0.05). To sum up the results above, it is clear that the effect of BSQDKT is more significant than BDKT on modulating immunosenescence, improving free radicals metabolism, anti-imparement of free radicals and improving function of cell membrane. This study provides an experimental basis for our concepts that zhengxujiayu (deficiency of vital energy complicated with blood stasis) is the main cause/mechanism of aging and fuzhenghuayu (kidney-tonifying spleen-invigorating blood stasis removing) method is probably the main route for delaying aging.
