米非司酮对卵巢癌细胞skov3耐药性表达的影响 被引量:1

摘要 目的:研究在阿霉素诱导卵巢癌细胞株skov3产生耐药性的过程中米非司酮所起的作用。方法:在阿霉素诱导skov3产生耐药过程同时使用不同浓度的米非司酮产生出各不同浓度的米非司酮与阿霉素诱导后的skov3细胞亚株,免疫组化SP法观察各细胞亚株P糖蛋白(P-glyco-protein,P-gp)表达情况;MTT法及annexin V-FITC/PI荧光染色流式细胞仪检测各细胞亚株死亡率及凋亡率;以罗丹明123(Rohdamin123,Rh123)作为荧光探针用流式细胞仪观察各细胞亚株胞内罗丹明含量;Western Blot法检测各细胞亚株的P-gp含量。结果:诱导过程中使用米非司酮使诱导后的skov3的P-gp表达降低;胞内罗丹明蓄积增加并可以提高同浓度阿霉素诱导后的细胞致死率及凋亡率。结论:米非司酮可以减慢卵巢癌耐药性的表达。 Objective:To study the effects of RU486 treated with adriamycin and to study the function and expression of muhidrug resistance in ovarian cancer cells line skov3. Methods: Immunohistochamical method was used to determine the expression of P - gp in cells line skov3 and other cells lines treated with different concentration RU486 and adriamycin. MTF and annexin V - FITC/PI method was used to study the apoptosis rate. Rohdamin 123 was used as fluorescent probe to observe the effects of every cell lines. Western Blot was used to determine the expression of P - gp. Results : The cell lines treated by RU486 can raise apoptosis rate and can increase the accumulation of Rohdamin 123 in cells and can decrease the expression of P - gp in cell lines. Conclusion: RU486 can decrease the function of multidrug resistance in cell lines treated with adriamycin.
