摘要 目的研制针对人隐孢子虫的特异性单克隆抗体。方法利用人隐孢子虫(C. hominis,)超声粉碎物免疫BALB/C小鼠,取脾细胞与SP2/0细胞融合,经3~5次有限稀释克隆化,获得7株持续分泌抗体的杂交瘤细胞株,经流式细胞术检测其反应性。结果获得7株单克隆抗体分别命名为JF1~JF7,对隐孢子虫胞内和胞外的检测无差异。结论所获得的单克隆抗体为临床对人隐孢子虫的诊断提供用力工具。 Objective To prepare specific moneclonal antibodies to Cryptosporidium hominis ( C hominis) . Methods Seven hybirdoma producing McAbs against occyst antigens of C hominis were developed by fusion of SP2/0 mouse myeloma cells and splecn cells from BALB/C mice immunized with sonicated occysts. Flowcytometry was used to assay the activity of McAbs. Results Seven McAbs obtained were named as JF1 - JF7 and there no differences were observed in extracellular and extracellular detection of Cryptosporidium hominis by flow cytometry. Conclusion The McAbs obtained will provide a valuable tool for in vivo or in vitro diagnosis of C hominis infection by flow cytometry.
