方格星虫多糖抗菌活性的初步研究 被引量:29

摘要 目的研究方格星虫多糖的抗菌活性。方法以方格星虫体壁作为试验材料,经沸水抽提、乙醇沉淀和sevage法去蛋白后得到粗多糖,再经G-50葡聚糖凝胶柱层析纯化。采用硫酸-苯酚法测定多糖的含量,以含不同浓度的方格星虫多糖(50,37.5,25,12.5mg/ml)培养基,分别接种金黄色葡萄球菌、大肠埃希菌和枯草杆菌培养12、24、36、48h,观察并记录其生长情况。结果不同方格星虫多糖浓度(25~50mg/ml)对三种试验菌的生长均有抑制作用,浓度越高抗菌活性越强。结论方格星虫多糖对金黄色葡萄球菌、大肠杆菌埃希菌、枯草杆菌有明显的抗菌活性。 Objective To explore the antibacterial activity of polysaeeharides contained in Sipunculus andus. Methods The polysaccharides contained in body wall of Sipunculus nudus were obtained. Crude polysaccharides were isolated by water boiling, ethanol precipitation and sevage method. Then they were purified hySephadex - G- 50 chromatography. The concentration of polysaccharide was deteetedby sulphuric acid phenol method. Staphylococcus aureus, escherichia coli and bacillus subtilis were exposed to different concentrations of 50, 37.5, 251, 12.5mg/ml of polysaccharides contained in Sipunculus nudus for 12,24,36,48 hour srespeetively . The inhibitory effects was observed on the nutrient medium. Results Three bacteria were restrained by different concentrations of polysaccharides contained in Sipunculus nudus , the antibacterial activity was increased in dose - dependent ways by pelysaccharides contained in Sipunculus nudus . Conclusion Polysaccharides contained in Sipunculus shows significant antibacterial activity on Staphylococcus aureus , escherichia coli and bacillus subdlis .
