高效液相色谱法测定血及腹腔组织中丝裂霉素浓度

Determination of mitomycin C in plasma and celiac tissue by high performance liquid chromatography method

目的 应用反相高效液相色谱方法（HPLC）测定血及腹腔组织中丝裂霉素C（MMC）含量。方法 血浆和组织液均用乙酸乙酯提取，利用反相高效液相色谱法测定组织中MMC。结果 MMC组织浓度线性范围为80～8000ng·mL^-1（r=0．99694），方法回收率为84％～96％。日内日间精密度（RSD％）为1．45％～14．10％，最低检测浓度为5ng·mL^-1，高于现有其他方法。结论 本方法简便、准确、灵敏，可用于科研和临床工作中血及组织中MMC浓度的快速检测。

Objective To determined the contents of mitomycin C （MMC） in plasma and celiac tissue by reversed-phase high performance liquid chromatography（RPHPLC） method. Methods Plasma and tissue suspension were extracted with ethyl acetate. MMC in plasma and celiac tissue was determined by RPHPLC. Results The linear range of MMC in tissue was from 80 ng . mL ^-1 to 8 000 ng. mL^-1（ r = 0.996 94）. The recovery was 84% - 96% , The relative standard deviation within a day and between days was 1.45% - 14.10 %. The minimum of quantitation was 5 ng . mL^-1. Conclusion This method is sensitive and simple and accurate, and could be used for determining the concentration of MMC in blood and tissue in clinic and scientific research.
