新生SD大鼠坐骨神经Wallerian变性规律及其S-100蛋白表达和雪旺细胞增殖研究 被引量:1

摘要 目的:了解新生SD大鼠周围神经Wallerian变性不同时间点S-100蛋白表达变化及对)旺细胞增殖的影响。方法:10d龄SD大鼠坐骨神经进行结扎预变性,分别于术后24h、3d、5d、7d和9d,进行)旺细胞培养和S-100免疫组织化学染色观察。结果:在损伤周围神经的近端发生可逆性变性,远端则发生永久性Wallerian变性。实验组)旺细胞增殖速率与S-100蛋白阳性染色率均优于对照组,7d组增殖能力达到高峰,S-100蛋白阳性染色率约51%~56%,与对照组比较差异有显著性。结论:新生SD大鼠周围神经受损后变性过程类似于经典的Wallerian变性,且能在变性中期获得活力较强及倍增时间较短的)旺细胞。 Objective: To study the S-100 expression change and the effect of reproductive activity on Schwann cells at different time points during Wallerian degeneration of the peripheral nerve of the neonatal SD rats. Methods: The sciatic nerves of the 10-day rats were ligated and pre-degenerated and the experimental result was analyzed with the methods of Schwann cells culture and S-100 protein was post- operatively observed in 24 hours, the 3th day, 5th day, 7th day and 9th day, respectively. Results: The reversible degeneration could take place at the proximal extremity of the damaged peripheral nerve, but the permanent Wallerian degeneration took place at the distal extremity. The reproductive rate of Schwann cells and the positive staining rate in the experimental group were better than that of the control group. The reproductive ability in the 7th day group reached the peak and the positive rate of S-100 protein was about 51%-56% in the 7th day. It had significant difference when compared to the control group. Conclusion: The degenerative process of damaged peripheral nerve of the neonatal SD rats is similar to the process of the typical Wallerian degeneration. The stronger reproductive activity and the shorter doubling time of Schwann cells can be obtained during the middle stage of Wallerian degeneration.
