摘要
目的观察淡豆豉提取物对心肌缺血的保护作用。方法将50只小鼠随机分为5组,分别为空白对照组、模型对照组、淡豆豉提取物小剂量(20g生药/kg)治疗组、淡豆豉提取物大剂量(40g生药/kg)治疗组及阳性药硝苯地平组(12mg/kg)。造模前治疗组灌胃给予不同剂量的淡豆豉提取物,连续7d,7d后腹腔注射脑垂体后叶素20U/kg,诱发心肌缺血。缺血20min后,观察各组心肌酶、自由基损伤指标的变化。结果模型对照组LDH、MDA明显较空白对照组升高,SOD、NO水平较空白对照组降低,淡豆豉提取物治疗组血清及心肌SOD、NO含量显著较模型对照组提高,LDH、CK、MDA明显较模型对照组降低。结论淡豆豉提取物对心肌缺血有一定的保护作用,其机制可能与抑制脂质过氧化反应,保护内皮依赖性松弛因子有关。
Objective To observe the protective effect of fermented soybean extraction on myocardial ischemia in mice. Methods 50 mice were randomly divided into 5 groups: blank control group, model controlgroup, low-dose treatment group with fermented soybean extraction(20 g/kg); high-dose treatment group with fermented soybean extraction(40 g/kg) and positive control group (nifedipine, 12 mg/kg). Before establishing the model, the mice in treatment groups were given with different doses of fermented soybean extraction by gavage for 7 days. Then pituitrin 20 U/kg was administrated via ip to induce myocardial ischemia for 20 minutes. The chang es of myocardial enzyme and free radical injury, parameters in every group were observed. Results Compard with those of blank control group, the levels of LDH and MDA in model group were increased, however, the levels of SOD and NO were decreased. Compared with those of model control group, the levels of SOD and NO both in serum and myocardium of mice in treatment groups were significantly increased, while the levels of LDH, CK and MDA were significantly decreased.Conclusion Fermented soybean extraction could protect mice from the myocardial ischemia, and the mechanism may be related to inhibiting lipid peroxidatic reaction and protecting endothelium-depending relaxing factor.
