摘要 目的研究BMP2对室管膜前下区(anterior subventricular zone,SVZa)神经干细胞分化为TH+(酷氨酸羟化酶)神经元的作用。方法体外培养SVZa神经干细胞,以0、0.1、1、10、20、100(ng/ml)浓度的BMP2分别诱导SVZa神经干细胞分化,通过免疫组化和流式细胞仪技术检测不同浓度下SVZa神经干细胞分化为TH+神经元的比例。结果0.1~100(ng/ml)组的TH+细胞比例均明显高于0ng/ml组,10ng/ml组达最高峰,各组间差异均有统计学意义(P<0.05)。结论BMP2可以促进SVZa神经干细胞向TH+神经元分化,可以明显提高分化比例。 Objective To study the role of BMP2 in SVZa NSCs differentiating into TH^+ neurons. Methods The neural stem cells of SVZa were separated and cultured in vitro. The neural stem cells of SVZa were induced by 0,0.1,1,10,20,103 (ng/ml ) concentrations of BMP2. The TH^+ neuronal rate of differention of the neural stem cells of SVZa was detected by immunohistochemistry and flow cytometry (FCM).Results BMP2 could promote TH^+ neuronal differentiation of neural stem cells of SVZa at the concentrations of 0.1 ~ 100ng/ml.The rate of TH^+ neuronal differentiation peaked when the concentration of BMP2 was 10ng/ml. As for portion of TH^+ neurons differentiation, there was significant difference between all groups by analysis of variance( P 〈 0.05). Conclusion BMP2 can promote the TH^+ neuronal differentiation of the neural stem cells of SVZa and the rate of TH^+ neuronal differentiation of neural stem cells of SVZa significantly as well.
