摘要
目的探讨多巴胺(DA)对多巴胺能神经母细胞瘤细胞SH-SY5Y的神经毒性作用及可能机制。方法应用MTT法检测DA对SH-SY5Y毒性作用,通过吉姆萨染色、Hoechst 33258标记、透射电镜、DNA ladder以及TUNEL检测DA诱导多巴胺能神经细胞凋亡形态结构及化学特征。结果MTT法检测到DA直接造成多巴胺能神经母细胞损伤,并呈浓度依赖性,有统计学意义(P<0.05);吉姆萨染色、Hoechst33258标记、FACS、电镜、DNA ladder以及TUNEL均检测到细胞凋亡形态及化学特征。结论DA在体外可以造成DA能神经母细胞SH-SY5Y损伤,可能是通过凋亡机制造成细胞丢失的。
Objective To explore the neurotoxicity of dopamine on dopaminergic neuroblastoma SH-SY5Y cells in vitro. Methods Human SH-SYS cells was used to study dopamine nerurotoxicity and analyze the mechanism of cell death by MTT, giems staining, Hoechst 33258, electron microscope, TUNEL, DNA ladder in vitro. Results Using MTT, giems staining, Hoechst33258, electron microscope, TUNEL, DNA ladder, we observed the dopaminergic neurons damage by dopamine, and detected the morphological and biochemical hallmarks of apoptosis. Conclusion The results suggest that dopamine can cause the degeneration of dopaminergic neuroblatoma SH-SYSY cells in vitro, and apoptosis may involve in the dopaminergic nerons death damaged by dopamine.
