铜绿假单胞菌诱导不同分化和活化状态的U937细胞分泌TNF-α和IL-8的研究 被引量:1

摘要 目的探讨铜绿假单胞菌诱导单核吞噬细胞产生细胞因子,以及细胞因子在宿主防御铜绿假单胞菌感染中的调节作用。方法应用佛波酯(PMA)刺激人单核白血病细胞系-U937细胞分化成巨噬细胞样细胞,对铜绿假单胞菌诱导不同分化和活化状态的U937细胞产生TNF-α和IL-8进行研究。结果铜绿假单胞菌不能诱导U937细胞产生TNF-α,但可诱导U937细胞产生IL-8及PMA分化的U937细胞产生TNF-α和IL-8;重组IL-1β可增强铜绿假单胞菌诱导PMA分化的U937细胞产生TNF-α和IL-8。结论TNF-α和IL-8的产生可能与U937细胞的分化状态有关,而且TNF-α和IL-8的产生可能存在着不同的诱导机制;IL-1β对TNF-α和IL-8的产生具有调节作用。 To investigate the effect of Pseudomonas aeruginosa to induce production of cytokines and the regulatory role of cytokines in the infection with this organism,the human monocytic leukemia cell line U937 was stimulated with phorbol myristate acetate(PMA)to differentiate into macrophage-like cells in different differentiated and activated states in order to explore the effect on the production of cytokines induced by P.aeruginosa and the regulatory role on the host defense against infection.The experimental results showed that P.aeruginosa could induce production of TNF-α in the PMA-differentiated U937 cells,and also induce production of IL-8 both in U937 cells and in PMA-differentiated U937 cells,but it failed to induce TNF-α production in U937 cells induced by P.aeruginosa.These data suggest that the production of TNFα and IL-8 induced by P.seruginosa may be related to the differentiated state of monocytes and different mechanisms exist in their production.
