色素上皮衍生因子在人白内障房水及晶状体上皮细胞中的表达

摘要 目的探讨色素上皮衍生因子(PEDF)在人白内障眼房水及晶状体上皮细胞(LEC)中的表达水平与年龄的关系。方法收集老年性白内障130例和先天性白内障15例。超声乳化术中取房水,酶联免疫吸附法(ELISA)检测房水PEDF浓度;选上述先天性白内障和老年性白内障高龄者各15例,术中取前囊膜标本用蛋白质免疫印迹法(Western Blot)检测LEC的PEDF表达。结果患眼房水PEDF平均浓度为(180.9±78.6)ng/ml,且与年龄均呈负相关(r=0.821,P<0.001);患眼LEC中存在PEDF的蛋白表达,且幼龄组的表达水平远高于高龄组。结论患眼LEC中存在PEDF活性,其房水及LEC中的PEDF水平均随年龄增加而显著下降,提示PEDF可能以旁分泌和自分泌的方式调节或(和)保护LEC。 Objective To study the relationship between age and PEDF in human cataractous aqueous humor and lens epithelium. Methods The PEDF concentration in the aqueous humor was measured by enzyme-linked immunosorbent assay in senile (130cases) and congenital (155cases) cataract patients who underwent cataract phacoemulsification extraction surgery. Anterior lens capsular specimens from senile (15cases) and congenital (15cases) cataract were collected for Western Blot with polycolnal antibodies specific to human PEDE Results The mean aqueous level of PEDF was ( 180. 9± 78.6) ng/ml, with negative linear correlation of PEDF level and age (r = 0. 821, P 〈 0. 001 ) . PEDF protein was detected in LECs, with lower level in senile group. Conclusion The PEDF levels in human aqueous humor and LECs decrease with aging. PEDF also ex- press in human LECs, which suggest PEDF regulate and/or protect LECs by paracrine and autocrine.
