Cathepsin B反义寡核苷酸对食管癌EC9706细胞中CB mRNA表达的影响

摘要 目的:探讨组织蛋白酶B(CB)反义寡核苷酸(ASODN)对CB mRNA表达的影响。方法:将食管癌EC9706细胞分5组培养,5组分别用设计合成的3条封闭CB不同基因位点的反义寡核苷酸(ASODN1、ASODN2、ASODN3)及1条无关寡核苷酸(N-ODN)转染,另1组不转染(空白对照组)。采用完整细胞原位杂交技术观察各组EC9706细胞中CB mRNA表达情况。结果:3条CB ASODNs转染的EC9706细胞中CB mRNA表达均下调,差异无统计学意义,但均明显低于N-ODN组及空白对照组。结论:CB ASODN可抑制食管癌EC9706细胞中CB mRNA表达,为临床预防食管癌等恶性肿瘤浸润转移奠定理论了基础。 Objective: To investigate the effects of cathepsin B antisense oligodexynucleotide (ASODN) on mRNA expression of cathepsin B in human esophageal cancer EC9706 cells. Methods: One non-sense oligodexynucletide (N-ODN) and three ASODNs against different cathepsin B gene sites were transfected into EC9706 cells, then the expression of cathepsin B mRNA in EC9706 cells was studied by in situ hybridization. Results.. The expression of cathepsin B mRNA could be inhibited by ASODNs. there was no significant difference between three ASODNs, but there was significant difference between ASODNs and N-ODN or control group. Conclusion. The expression of cathepsin B mRNA in EC9706 cells could be inhibited by ASODNs,which provides the basic theory in preventing the invasion anti metastasis of esophageal carcinoma.
