摘要
从呼和浩特市周边地区采集的新鲜山羊奶,通过离心脱脂,调节等电点沉淀蛋白,对所得沉淀物进行DEAE离子交换层析,分离出α-酪蛋白和β-酪蛋白,并且β-酪蛋白被完全纯化出来。建立ELISA双夹心方法对β-酪蛋白的含量进行测定,该法可直接用于培养液中β-酪蛋白的定量分析。
To purification of goat β-casein and prepare its antiserum for ELISA method of goat β-casein research.. Goat β-casein was purified with DEAE anion exchange chromatography. It was used to immunize rabbits to acquire anti-serum. The anti-serum was analyzed with ELISA method. The results of ELISA showed the generation of high titer rabbit antibodies. Polyclonal antibodies to goatβ-casein was developed successfully. Double antibody sandwich ELISA were carried out in this study. By using of the chessboard titration method, the working concentrations of coated antibody, reactive first antibody and HRP-labeled second antibody were selected. Reference standard curve is in a hnear correlation pattern.The improved antibody sandwich ELISA is good at direct quantification of β-casein.
