黄芪、三七对下肢缺血患者骨髓干细胞体外分化作用的影响 被引量:3

摘要 目的探讨黄芪、三七对骨髓干细胞体外分化作用的影响。方法常规采集下肢缺血患者骨髓血,密度梯度离心法分离骨髓单个核细胞,在黄芪、三七不同浓度条件下贴壁扩增细胞。3周后行镜下细胞的形态学观察,流式细胞仪检测CD34+细胞的百分比。结果细胞呈梭形,束状排列,间杂有少量圆形细胞。与对照组比较,黄芪中剂量和低剂量组、三七中剂量组和高剂量组CD34+细胞的百分比均显著增加,差异有显著性意义(P<0.05或P<0.01)。结论黄芪、三七有促进骨髓干细胞向血管内皮前体细胞(EPC)转化和增殖的作用。 Objective To explore the effect of herbal medicines Sanqi (Radix Notoginseng) and Huangqi (Radix Hedysari) on the differentiation in vitro of stem cells. Methods The marrow blood of the patient with isehemia of lower limbs was collected routinely. The density gradient centrifugation was adopted to separate the single nuclear cells, and the cells were amplified by under different concentration. Three weeks later, the morphological changes were observed. The flow eytometer (FCM) was used to test the percentage of CD34^+ cells. Results The cells was shaped in fusiform and arranged in bunchy order with few circular cells mixed. The percentage of CD34^+ cells was significantly increased in the Huangqi medium dosage group and Huangqi small dosage group and Sanqi medium dosage group, Sanqi large dosage group(P〈0. 05 or P〈0.01). Conclusion Huangqi and Sanqi have the function of promoting the transformation and proliferation of stem cell to vascular endothelia precursor cells (EPC).
