瘦素、趋化因子受体-3和钙结合蛋白-A12基因在热痹中的表达

The Expression of Leptin,Chemokine (C-C motif) Receptor 3 and S100 Calcium Binding Protein A12 Gene in Pyretic Arthralgia

摘要 目的运用基因表达谱芯片技术分析痹病寒热证型的基因表达差异,探索炎性反应相关基因在痹病分型中的表达情况。方法选取4例寒痹患者(C组)与3例热痹患者(H组)的滑膜组织标本,制备标记探针,采用美国Affymetrix公司提供的GeneChip Human Genome U133Plus2.0芯片进行杂交与扫描程序。结果筛选出P<0.05且H/C差异表达变化超过2倍的差异表达基因,鉴别出3条炎性反应相关基因,即瘦素(leptin,LEP)、趋化因子(C-C基序)受体-3[chemokine(C-C motif)receptor3]和钙结合蛋白-A12(S100calcium binding protein A12)基因,它们均在H组中高表达。结论LEP、CCR3、S100A12基因与热痹存在高度关联,其编码产物可能是热痹的活性分子标志物。 Objective Microarray analysis was used to evaluate the difference in gene expression in synovial tissue samples obtained from patients with arthromyodynia, and to explore the role of inflammatory reaction-related gene in pyretic arthralgia. Method Four samples were obtained from the patients with cold hypochondrium protrusion and three from patients with pyretic arthralgia during arthroscopic knee surgery. The probe was prepared and then hybridization and scanning were conducted by using GeneChip Human Genome U133 Plus 2. 0. Result The differential genes with P value 〈 0. 05 and a two-fold change in H/C expression were selected. From these genes, 3 inflammation-related genes, i. e. leptin ( Lep), chemokine ( C-C motif) and S100 calcium binding protein A12, were identified and they all had high expression in group H. Conclusion This experiment confirmed that leptin, chemokine (C-C motif) receptor 3 and SIO0 calcium binding protein A12 gene are closely related to pyretic arthralgia and their encoding products may be bioactive marking molecules of pyretic arthralgia.
